The MotoGP titleholder emerged victorious in the 10-lap contest, finishing ahead of Andrea Iannone, who had begun the race from the top starting spot.
Marc Marquez secured a spot on the podium after Nicolo Bulega crashed in the final corner of the race.
Bagnaia managed to pass Iannone, who started in pole position, during the first lap and stayed in the lead. Although Iannone fought hard, he ultimately had to accept second place.
The highlight of World Ducati Week, the Race of Champions, takes place at the Misano circuit in Italy to honor the Italian motorcycle maker.
The event united Ducati's competitors from MotoGP, World Superbikes, and British Superbikes.
The Panigale V4 was revealed earlier and utilized by racers during the competition.
